Gonadotropin-releasing hormone (GnRH) is a neuropeptide with various roles in regulating reproductive systems. However, expression of GnRH-receptor in human breast carcinomas has not been studied. In this study, GnRH-A receptor mRNA and protein expressions in human breast carcinomas were examined. Using a polymerase chain reaction (PCR) method, GnRH-A receptor mRNA was detected in 13 of 21 (62%) human breast carcinoma tissues, but was not detected in 18 breast carcinoma tissues in which human epidermal growth factor receptor 2 (HER2) was amplified. In contrast, GnRH-A receptor mRNA was not detected in the normal breast tissues. Quantitative real-time reverse transcription-PCR analysis showed that GnRH-A receptor mRNA levels in human breast carcinoma tissue were higher than those in the normal breast tissues. Immunohistochemical analysis demonstrated that GnRH-A receptor was expressed in both luminal and/or myoepithelial cells in human breast carcinoma tissues, and that the GnRH-A receptor protein expression level was also upregulated in human breast carcinomas as compared with the normal breast tissues.
